I wasn't sure what new twists 3rd edition has added to the equation so I
thought I would ask. What does it take in Shadowrun to actually kill a
vampire?

(I'm considering adding a Vampire hunting story to the mix and just wanted
to know what the mechanics are before working out the plot.)

Hmm, interesting question. I don't have the Third Edition rules, but as
someone who's been running some vampires here on the list my idea is that it
takes something like what White Wolf calls "aggravated damage". My conception
of vampires is of beings that simply can't be killed by most kinds of mundane
means. Shoot them with enough normal bullets to grind them into hamburger,
and they can still regenerate - it just might take awhile. Inflict magical
damage, or use fire (flame thrower, willie-pete, etc.) and you may be getting
somewhere.

For the record, the little scene I posted of Thelienista is my conception of
one vampire trying to perfect a weapon skill under the tutelage of another.
Thel got hit with a D+ attack that drove her ribs into the contents of her
ribcage, and then eviscerated and had her throat cut. On the assumption that
she hadn't been "starving" up to that point, her wounds were still able to
heal on order of a minute, although I depicted her being "hungry" afterward.
Mind you, that was mundane, not aggravated, damage she suffered.

If you're interested in doing something with the Kindred, maybe we could
collaborate. I've been looking for ways to get them back into the mix, and
not having a dreadful amount of inspiration as of late. I've got some ideas
about how the Kindred might be (dis?)organized in the Shadowrun setting...
